Albert stain is useful for the identification of Corynebacterium diphtheriae by staining it using its metachromatic granules and cytoplasm. The mentioned bacterium is identified as its body appears to be green in color and the granules has blue black appearance. Using Albert Stain A Solution is a special type of staining technique in which different staining colors can also be used. By using polychrome methylene blue, appearance of bacteria is blue in color whereas granules appear to be violet. Albert's A Solution includes ethyl alcohol, glacial acetic acid, Toluidine blue and malachite green.
